Triple-Negative Breast Cancers with Amplification of JAK2 at the 9p24 Locus Demonstrate JAK2-Specific Dependence
Targeted next-generation sequencing of postchemotherapy triple-negative breast cancers (TNBC) identified a group of 9p24-amplified tumors, which contained focal amplification of the Janus kinase 2 (JAK2) gene. These patients had markedly inferior recurrence-free and overall survival compared to patients with TNBC without JAK2 amplification. [Sci Transl Med] Abstract

Cartilage Oligomeric Matrix Protein Contributes to the Development and Metastasis of Breast Cancer
In vitro experiments confirmed that cartilage oligomeric matrix protein (COMP)-expressing cells had a more invasive phenotype, which could in part be attributed to an upregulation of matrix metalloprotease-9. Furthermore, microarray analyses of gene expression in tumors formed in vivo showed that COMP expression induced higher expression of genes protecting against endoplasmic reticulum stress. [Oncogene]

miR-155 Downregulates ErbB2 and Suppresses ErbB2-Induced Malignant Transformation of Breast Epithelial Cells
Scientists found that ErbB2 expression was inversely correlated with the level of miR-155, a well-documented oncogenic miRNA, in ErbB2-positive breast tumors. They further determined that miR-155 potently suppresses ErbB2 in breast cancer cells. [Oncogene] Abstract

Chemotherapy-Induced Uridine Diphosphate Release Promotes Breast Cancer Metastasis through P2Y6 Activation
The authors demonstrated that UDP/P2Y6 signaling facilitates breast cancer metastasis both in vitro and in vivo. They found that P2Y6 is not only aberrantly expressed and mutated in most tumor types, but also highly correlated with poor prognosis in breast cancer patients. [Oncotarget] Full Article

GPER Signaling in Both Cancer-Associated Fibroblasts and Breast Cancer Cells Mediates a Feedforward IL1β/IL1R1 Response
Investigators demonstrated that signaling mediated by the G protein estrogen receptor (GPER) triggers IL1β and IL1R1 expression in cancer-associated fibroblasts and breast cancer cells, respectively. [Sci Rep] Full Article

Bioorthogonal Two-Component Drug Delivery in HER2(+) Breast Cancer Mouse Models
Researchers developed a two-component, two-step, pre-targeting drug delivery system integrated with image guidance to circumvent these issues. In this strategy, HER2 receptors were pre-labeled with a functionalized trastuzumab antibody followed by the delivery of drug-loaded nanocarriers. [Sci Rep] Full Article

A Novel EGR-1 Dependent Mechanism for YB-1 Modulation of Paclitaxel Response in a Triple Negative Breast Cancer Cell Line
Researchers aimed to determine mechanisms by which Y-box binding protein 1 (YB-1) confers paclitaxel (PTX) resistance. They generated isogenic triple negative breast cancer cell lines that differed by YB-1 levels and treated these with PTX. [Int J Cancer] Abstract

Parthenolide and DMAPT Exert Cytotoxic Effects on Breast Cancer Stem-Like Cells by Inducing Oxidative Stress, Mitochondrial Dysfunction and Necrosis
Researchers report the production of mammospheres from three lines of triple-negative breast cancers cells and demonstrated that both parthenolide and its soluble analog dimethylaminoparthenolide (DMAPT) suppressed this production and induced cytotoxic effects in breast cancer stem-like cells, derived from dissociation of mammospheres. HDAC8 Inhibition Blocks SMC3 Deacetylation and Delays Cell Cycle Progression without Affecting Cohesin-Dependent Transcription in MCF7 Cancer Cells
Scientists blocked deacetylation of ac-SMC3 using an HDAC8-specific inhibitor PCI-34051 in MCF7 breast cancer cells, and examined the effects on transcription of cohesin-dependent genes that respond to estrogen. [J Biol Chem] Abstract | Full Article
