How Breast Cancer Usurps the Powers of Mammary Stem Cells
Scientists have found that mammary stem cells associated with the pregnant mammary gland are related to stem cells found in breast cancer. They specifically identified a key molecular pathway associated with aggressive breast cancers that is also required for mammary stem cells to promote lactation development during pregnancy. NSAID Use Reduces Breast Cancer Recurrence in Overweight and Obese Women: Role of Prostaglandin-Aromatase Interactions
Researchers conducted in vitro studies that utilized sera from obese and normal-weight patients with breast cancer. Conditioned media (CM) from macrophage/preadipocyte cocultures exposed to sera from obese patients stimulated greater breast cancer cell estrogen receptor α activity, proliferation, and migration compared with sera from normal-weight patients, and these differences were eliminated or reduced by the addition of an aromatase inhibitor during CM generation. Oncogenic Suppression of Apoptosis Uncovers a Rac1/JNK Proliferation Pathway Activated by Loss of Par3
Scientists show that loss of Par3 in mammary epithelial cells promotes apoptosis, and that oncogenic Notch overcomes the apoptotic signal to reveal an unexpected pro-proliferative role for loss of Par3 in mammary tumors. [Oncogene] Abstract

Geranylgeranylation Signals to the Hippo Pathway for Breast Cancer Cell Proliferation and Migration

Modeling Luminal Breast Cancer Heterogeneity: Combination Therapy to Suppress a Hormone Receptor-Negative, Cytokeratin 5-Positive Subpopulation in Luminal Disease
To address the relationships between estrogen (ER)+ progesterone (PR)+ cytokine 5 (CK5) and ERPRCK5+ cells in Luminal cancers and tightly control their ratios scientists generated isogenic pure Luminal and pure Luminobasal cells from the same parental Luminal human breast cancer cell line. Impaired Cell Death and Mammary Gland Involution in the Absence of Dock1 and Rac1 Signaling
Researchers generated two mouse models with conditional inactivation of dedicator of cytokinesis 1 (Dock1) and ras-related C3 botulinum toxin substrate 1 (Rac1) and revealed that the expression of these genes is not essential in the mammary gland during puberty, pregnancy and lactation. TNF Inhibitor Therapy and Risk of Breast Cancer Recurrence in Patients with Rheumatoid Arthritis: A Nationwide Cohort Study

Effect of Bisphosphonate Use on Risk of Postmenopausal Breast Cancer
Observational studies have suggested a possible protective effect of bisphosphonates on breast cancer, but the effect of bisphosphonate use on risk of breast cancer has not been tested in randomized trials. Investigators assessed the relationship of postmenopausal breast cancer incidence and bisphosphonate use using data from two randomized (1:1), double-blind, placebo-controlled trials.
