Johns Hopkins Scientists Discover New Therapeutic Target for Some Breast Cancers  ShareThis
A protein that pumps calcium out of cells also moonlights as a signal to get massive quantities of calcium to flow in, according to scientists. Their discovery of this surprisingly opposite function, highlights the link between calcium and cancer and holds the promise of a new therapeutic target for certain breast cancers. [Press release from the Johns Hopkins University School of Medicine discussing online prepublication in Cell]

LABORATORY RESEARCH

Store-Independent Activation of Orai1 by SPCA2 in Mammary Tumors
Researchers show that an isoform of secretory pathway Ca2+-ATPase, SPCA2, is upregulated in breast cancer-derived cells and human breast tumors, and suppression of SPCA2 attenuates basal Ca2+ levels and tumorigenicity. [Cell]

miR-378* Mediates Metabolic Shift in Breast Cancer Cells via the PGC-1Beta/ERRGamma Transcriptional Pathway
Here, researchers show that miR-378* expression is regulated by ERBB2 and induces a metabolic shift in breast cancer cells. [Cell Metab]

Cancer Stem Cells From Human Breast Tumors are Involved in Spontaneous Metastases in Orthotopic Mouse Models
To examine the role of breast cancer stem cells in metastasis, researchers generated human-in-mouse breast cancer orthotopic models using patient tumor specimens, labeled with optical reporter fusion genes. [Proc Natl Acad Sci U S A]

Downregulation of Autophagy by Bcl-2 Promotes MCF7 Breast Cancer Cell Growth Independent of Its Inhibition of Apoptosis
Here, by using a loss-of-function mutagenesis study, researchers show that Bcl-2-mediated antagonism of autophagy has a critical role in enhancing the tumorigenic properties of MCF7 breast cancer cells independent of its anti-apoptosis activity. [Cell Death Differ]

Int6 Regulates Both Proteasomal Degradation and Translation Initiation and is Critical for Proper Formation of Acini by Human Mammary Epithelium
Researchers found that, repressing INT6 expression induced transformed properties in normal human mammary epithelium (MCF10A); in contrast, Int6 silencing induced apoptosis in HeLa cells. [Oncogene]

The Normal Mammary Microenvironment Suppresses the Tumorigenic Phenotype of Mouse Mammary Tumor Virus-Neu-Transformed Mammary Tumor Cells
The results indicate that signals emanating from a normal mammary microenvironment, comprised of stromal, epithelial and host-mediated signals, combine to suppress the cancer phenotype during glandular regeneration. [Oncogene]

Cancer Cell Stiffness: Integrated Roles of Three-Dimensional Matrix Stiffness and Transforming Potential
Given the current knowledge base of breast cancer mechanobiology, the findings suggest that there may be a positive relationship among intracellular stiffness sensitivity, cell motility, and perturbed mechanotransduction in breast cancer. [Biophys J]

CLINICAL RESEARCH

High Expression of Sphingosine 1-Phosphate Receptors, S1P1 and S1P3, Sphingosine Kinase 1, and Extracellular Signal-Regulated Kinase-1/2 Is Associated with Development of Tamoxifen Resistance in Estrogen Receptor-Positive Breast Cancer Patients
A cohort of 304 estrogen receptor (ER)-positive breast cancer patients was used to investigate the prognostic significance of sphingosine 1-phosphate (S1P) receptors 1, 2, and 3 (ie, S1P1, S1P2, and S1P3), SK1, and extracellular signal-regulated kinase 1/2 expression levels. [Am J Pathol]

Zoledronic Acid Prevents Bone Loss in Premenopausal Women with Early Breast Cancer Undergoing Adjuvant Chemotherapy: A Phase III Trial of the Korean Cancer Study Group (KCSG-BR06-01)
The purpose of this study was to determine whether zoledronic acid could prevent bone loss in premenopausal women undergoing adjuvant chemotherapy for breast cancer. [Breast Cancer Res Treat]
